The SUN-domain proteins are associated with both nuclear lamins and chromatin and cross the inner nuclear membrane. Recently, the LINC complex has been described for its role LInking the Nucleus to the Cytoskeleton and may play an important role in signaling and structural integrity. The LINC complex consists of large isoforms of nesprin-1 and -2 facing the cytoplasm. The nesprins are tethered to the outer nuclear membrane by interaction with SUN proteins. The LINC complex has a core 6:6 structure in which KASH-binding induces head-to-head interactions between SUN trimers, suggesting force transduction between cytoskeletal and nuclear components through branched LINC complex networks. The LINC complex is made of Sad1/Unc84 (SUN) and Klarsicht/Anc1/Syne1 homology (KASH) proteins which have been recently characterized in plants.
